摘要
目的讨论肿瘤坏死因子α(TNF-α)和基质金属蛋白酶-3(MMP-3)在子宫内膜异位症(EMs)中的表达和发病的关系。方法以EMs患者20例作研究组,分别取其术前清晨空腹静脉血3mL和在位与异位内膜,以子宫肌瘤患者20例的血清和内膜为对照组。内膜中的MMP-3采用免疫组化方法测定。血清中TNF-α和MMP-3的含量采用ELISA法检测。结果MMP-3、TNF-α在血清中的含量在实验组明显高于对照组(P<0.01)。MMP-3主要定位于子宫内膜组织的间质细胞。在异位内膜中,其阳性染色强度均较强,以(+)~(++)为主,呈棕黄色或棕褐色,明显强于正常子宫内膜(P<0.05),差异有统计学意义。MMP-3在正常子宫内膜组中增殖期的表达强于分泌期(P<0.05)。结论MMP-3在EMs患者异位内膜和在位内膜中的高表达,促进了内膜的侵袭、浸润。TNF-α和MMP-3在EMs患者血清中的含量升高,在黏附的同时促进新生毛细血管的形成,加速了EMs的发生发展。
Objective To investigate the expression and significanceof the TNF-α and MMP- 3 in patients with endometriosis (EMs). Methods 20 cases with EMs ( research group ) and 20 cases with the uterine fibroid patients( control group) were selected. MMP-3 in endometrium was measured by immunohistochemistry method. Serum TNF- α and MMP- 3 were measured by ELISA method. Results MMP- 3 positioned mainly in the endometrial stromal ceils of endometrium. The serum TNF - α and MMP - 3 , in research group were significantly higher than those in control group( P 〈 0.05 ). Expression of MMP -3 in normal endometrial proliferation was significantly stronger than that in secretory phase (P 〈 0.05). Conclusion High expression of MMP - 3 in patients with EMs enhance and promote the transfer of the endometrium and infiltration. Rising of the serum TNF -α and MMP - 3 promote new capillary formation and accelerated the development of EMs.
