Effects of norepinephrine on the activation of LPS-induced macrophages

摘要 目的检测去甲肾上腺素(norepinephrine,NE)对脂多糖(lipopolysaccharide,LPS)诱导的腹腔巨噬细胞活化的影响,并探讨其可能的分子机制。方法收集大鼠腹腔巨噬细胞,分别用不同浓度(2~100ng/ml)NE预处理1h或以10ng/mlNE分别预处理0.5~5h后,再用LPS刺激1h,分别检测巨噬细胞吞噬功能和分泌TNF-α能力;RT-PCR法检测其CD14、清道夫受体(scavenger receptor,SR)和TLR4表达水平。结果低浓度(2~10ng/ml)NE显著促进LPS诱导的巨噬细胞吞噬和分泌TNF-α,而更高浓度(50~100ng/ml)对此无作用;时效作用方面,NE处理1h对LPS诱导的TNF-α分泌促进作用最明显。同时,低浓度(2~10ng/ml)NE促进巨噬细胞CD14和SR的表达,更高浓度对其表达无影响;各浓度NE对TLR4表达水平无影响。结论一定浓度NE可增强LPS对巨噬细胞的活化,并且此过程与巨噬细胞CD14和SR表达水平升高有关。 Objective To observe the effects of norepinephrine on the activation of macrophages induced by LPS and to explore its possible molecular mechanisms. Methods The peritoneal macrophages collected from male SD rats were pretreated by norepinephrine at different concentrations for 1 h, or norepinephrine at the dose of 10 ng/ml for a period ranging from 0.5 h to 5 h, and then treated by LPS for another 1 h. The phagocytic capacities and TNF-α secretion abilities of macrophages were measured. The CD14, SR, and TLR4 mRNA expression levels were evaluated by semi-quantitative RT-PCR. Results Norepinephrine at low concentrations (2 ng/ml -10 ng/ml) predominantly enhanced the phagocytic capacities and the TNF-α secretion abilities of peritoneal macrophages induced by LPS, but it had no effects on these functions at higher concentrations (50 ng/ml - 100 ng/ml). Besides, norepinephrine at low concentrations improved CD14 and SR mRNA expression levels of peritoneal maerophages, but it had no effects at higher concentrations. Further more, norepinephrine had no effects on TLR4 mRNA expression at the observed concentrations. Conclusion Norepinephrine at a suitable concentration can enhance the activation of macrophages induced by LPS, which may be associated with the elevated expressions of CD14 and SR mRNA.
