三羟异黄酮对氧化损伤内皮细胞保护作用的实验研究 被引量:1

Experimental Research on Protective Effect of Phytoestrogen on Human Umbilical Vein Endothelial Cells from Oxidative Injury

摘要 目的研究植物雌激素的有效成分三羟异黄酮对胆固醇致人脐静脉内皮细胞(HUVEC)损伤的保护作用,探讨植物雌激素的抗氧化功能。方法体外培养HUVEC,将细胞分为5组:正常对照组、胆固醇损伤组(胆固醇50mg/L+终浓度为10μmol/L的CuSO4溶液)、胆固醇损伤的同时加入三羟异黄酮不同剂量,GST低剂量组(22.5μmol/L)、中剂量组(45μmol/L)和高剂量组(90μmol/L)。培养结束后,测定上清培养液中谷胱甘肽过氧化物酶(GSH-Px)活力、丙二醛(MDA)含量以及一氧化氮(NO)生成量。结果胆固醇损伤组的GSH-Px活力和NO生成量明显下降,MDA含量升高,与正常对照组和GST不同剂量组比较差异有显著性(P<0.05),而正常对照组和GST不同剂量组比较差异无显著性(P>0.05)。结论胆固醇可以诱导内皮细胞损伤,三羟异黄酮可能有阻止胆固醇对HUVEC的损伤作用。 Objective To study the protective effect of Genistein on human umbilical vein endothelial ceils (HUVEC) which were damaged by cholesterin and explore its anti - oxidiration. Methods HUVEC cells were cultured in vitro and divided into 5 groups: I. normal control group;2. Cholesterin damage group (cholesterin 50mg/L + 10μmol / L CuSO4 ] ; 3. Low - dose GST group (22.51μmol/L); 4. medium - dose GST group (45μmol/L) ; 5. high - dose GST group [90μmol/L) ; After cultured , GSH -Px activity, MDA content and the concentrition of NO in HUVEC culture medium were measured. Results These results showed that GSH - Px activity and the concentration of NO decreased significantly in Cholesterin damaged group and MDA content increased, Compared with control group and different doses of GST group ( P 〈 0.05 ) , and there was no significant difference between control group and different doses of GST group ( P 〉0. 05 ). Conclusion Cholesterin could induce injury of HUVECS. GST may prevent HUVECS from cholesterin induced the injury. l
