摘要
目的:建立芹菜素的高效液相色谱检测方法,研究芹菜素与蛋白的相互作用。方法:采用C8柱(250mm×4.6mm,5μm)进行色谱分析,梯度洗脱,检测波长337nm,流速1.0mL·min-1;同时,结合平衡透析法,研究了芹菜素与蛋白的相互作用。结果:在2min之内实现了芹菜素的快速检测,并且在37℃的水浴恒温培养4h后,芹菜素与蛋白发生相互作用,最大结合量为5.55,结合常数为0.17(mg·L-1)-1。结论:本法可以快速准确地检测芹菜素,并可研究其与蛋白的相互作用。
Objective: To establish an HPLC method for the determination of apigenin and investigate its interaction with human serum albumin (HSA). Methods:Apigenin was detected by high performance liquid chromatography with Cs column (250 mm×4. 6 mm,5 μm) and gradient elution under the detection wavelength of 337 nm and the rate of 1.0 mL·min^-1. Furthermore,its interaction with human serum albumin was investigated by equilibrium dialysis. Results:Apigenin was immediately detected in 2 min and a strong interaction between these two substances was found after 4 -hour culture in 37 ℃ water bath,with a binding constant of 0. 17 (mg · L^-1) -1 and the maximum binding amount to 5.55. Conclusion:The method is accurate, sensitive for the determination of apigenin, and can be used to investigate the interaction of human serum albumin with other pharmaceuticals.
