摘要
采用共混法制备壳聚糖/羟基磷灰石复合膜。膜物理性能测试表明,水相接触角由壳聚糖的103°下降到复合膜的57°,吸水率有所上升。将材料浸入培养基中,能吸附大量Ca2+,在表面形成一层低结晶态羟基磷灰石,表面变得粗糙。分别在壳聚糖和壳聚糖/羟基磷灰石复合膜表面培养骨髓间充质干细胞表明,经共混后的膜有利于骨髓间充质干细胞的生长贴附,在材料上细胞经诱导后,共混膜组碱性磷酸酶表达在培养12 d到达10.1,与壳聚糖组的1.6比较,具有显著差异(P<0.01)。上述结果表明,壳聚糖/羟基磷灰石膜具有良好的生物相容性,有利于细胞的增殖和分化,在骨移植和骨替代方面具进一步研究应用的价值。
Compound membranes of chitosan/hydroxyapatite were prepared by blending. The physical performante showed that the air-water contact angles decreased from ehitosan's 103°to chitosan/hydroxyapatite's 57°and the water adsorption rate increased slightly. When immersed into culture medium, the materials adsorbed Ca^2+ , and low crystalline hydroxyapatite deposited on the surface of the membranes. Chitosan/hydroxyapatite compound membranes could enhance the attachment and proliferation of mescenchymal stem cells (MSCs). After 12 days' induction on the materials, the alkaline phosphatase(ALP) activity value of MSCs on the compound membrane was 10. 1, being much higher than 1.6 on chitosan membrane(P〈0.01). All these results indicate that ehitosan does not have very good affinity for MSCs, but the bioeompatibility of chitosan can be apparently enhanced after mixing with hydroxyapatite. The compound membrane stimulates MSCs to differentiate into osteoblasts and it may be a good potential material for bone substitution.
