ESA刺激华支睾吸虫感染小鼠脾细胞分泌Th1/Th2的动态观察 被引量:7

Th1/Th2 cytokine profile in spleen mononuclear cells of mice infected with Clonorchis sinensis stimulated by excretory-secretory antigen

摘要 目的观察华支睾吸虫分泌排泄抗原(ESA)刺激感染小鼠脾脏单个核细胞Th1/Th2细胞因子的动态变化。方法双抗体夹心ELISA法检测华支睾吸虫感染后不同时间小鼠脾细胞培养上清中IFN-γ、IL-4的动态变化。结果自感染后1w起IFN-γ的表达量开始上升,第4w达高峰(P<0.05),此后开始下降,至第12w水平接近正常。与IFN-γ不同,IL-4的表达自感染后第1w起持续升高,直至观察结束的12w(P<0.05)。结论初步结果提示华支睾吸虫感染后急性期转向慢性期的过程中Th1/Th2细胞因子的表达经历了由Th1优势应答向Th2优势应答的转变。 Objective To investigate the dynamics of both Th1 and Th2 cytokines in spleen mononuclear cells of mice infected with Clonorchis sinensis stimulated by excretory-secretory antigen (ESA) and to analyze its role in clonorchiasis. Methods The Sandwich ELISA was used to measure the levels of IFN-γ and IL-4 in culture supematants of mouse splenocytes stimulated by ESA at different time points post-infection. Results The level of IFN-γ increased significantly during the acute stage and declined thereafter,with its peak at week 4 post-infection (P〈0.05). Whereas IL-4 production was significandy getting higher (P〈0.05) throughout the infection observed. Conclusion The investigation suggested that the preferential Thl cytokine response during acute stage of infection switched to Th2 type response in chronic stage.
