摘要
目的以养殖鲟鱼软骨为原料,提取硫酸软骨素、筛选提取工艺条件。方法用稀碱-盐解-酶解法提取硫酸软骨素,并对各工艺中影响提取效果的不同因素进行正交实验。结果碱提取最佳工艺组合为:碱浓度5%,料液比1∶6,碱提取时间3h;盐解最佳工艺组合为:盐浓度4%,盐解温度90℃,盐解时间20min;酶解最佳工艺组合为:酶浓度0.4%,酶解温度52℃,酶解时间2.5h。结论采用上述组合提取硫酸软骨素的得率为31.56%,显著高于(P<0.05)传统工艺的25.86%。
Purpose The chondoitin sulfate was isolated from cartilage of cultured Chinese sturgeon through a method of dilute alkali-enzyme hydrolysis. The extraction technique of ehondroitin sulfate was improved. Methods The Chondoitin sulfate was isolated through a method of dilute alkali-enzyme hydrolysis. Resuits To extract the chondroitin sulfate from sturgeon cartilage in dilate alkali, the best combination of extracting technique was : the 5 % alkali concentration, the 1 : 6 weight proportion, and the time of 3 hours. To extract the ehondroitin sulfate from sturgeon cartilage in dilute salt, the best combination was the 4 % salt concentra- tion, the temperature of 90, and the time of 20 minutes. In enzymeolysis, the best combination was 0.4 % enzyme concentration, the temperature of 52 ℃, and the time of 2.5 hours. Conclusion The yield of chondroitin sulfate increased significantly ( P 〈 0.05) after the optimization of the technique.
