神经生长因子在不同条件下的活性研究

摘要 目的研究各种因素对神经生长因子(NGF)生物活性及免疫原性的影响。方法把天然NGF在不同浓度的变性剂、去污剂、氧化还原剂及其配伍中或在不同pH值、不同温度下进行孵育处理,测定其生物活性和免疫原性的变化。结果NGF在8mol/L尿素中孵育过夜后生物活性下降10倍,在浓度低于5mol/L的尿素中孵育过夜后生物活性基本不变;十二烷基硫酸钠和巯基乙醇对NGF生物活性破坏较大,且尿素可增加巯基乙醇的破坏作用;还原型谷胱甘肽和氧化型谷胱甘肽配合使用对NGF生物活性的影响较小;NGF对温度敏感,在60℃孵育1h后活性降低约30倍,在37℃孵育48h生物活性开始下降;NGF在酸、碱中稳定。生物活性降低的NGF免疫原性未见明显改变。结论NGF的生物活性易受多种因素影响,但其免疫原性较稳定。 Purpose To study on the effect of several factors on the bioactivity and immunogenicity of natural nerve growth factor (NGF). Methods Natural NGF was treated by incubating under different conditions, including in denaturant of different concentrations, in detergent, in reduetant-oxidant and their combinations, or at different pH and temperatures. After removing the influencing factors, changes of the bioactivity and immunogenicity of the treated NGF were determined. Results Incubating in 8 mol/L urea resulted in 10-fold decrease of the bioactivity of NGF. Incubating in urea of 5 mol/L or lower did not change the bioactivity of NGF. SDS and mereaptoethanol could result in significant decrease of the bioactivity of NGF, and urea had a synergistic effect with mereaptoethanol. Incubating in the mixture of GSH and GSSG did not cause obvious decrease of the bioactivity of NGF. The bioactivity of NGF was sensitive to high temperature, and incubating at 60 ℃ for one hour resulted in about 30-fold decrease of the bioactivity. However NGF began to lose its bioactivity after being incubated at 37 ℃ for more than 48 hours;NGF was stable under acid and basic conditions. NGF with lowered bioactivity showed little change in the immunogenicity. Conclusion The bioactivity of NGF can be affected by many factors, but its immunogenicity is relatively stable.
