摘要
目的探讨新西兰大白兔角膜碱烧伤后及同种异体角膜移植术后角膜新生淋巴管(CL)上VEGFR3、LYVE-1及CD4的表达及意义,从而进一步证实角膜碱烧伤和高危角膜移植后角膜新生淋巴管的存在及与排斥反应的关系。方法饲养96只健康新西兰大白兔,将其随机分为5组。空白对照组24只,选取实验1组24只右眼,不做手术;实验1组24只,左眼制作角膜碱烧伤模型;实验2组24只,左眼进行钻取自体角膜后再缝合;实验3组24只,左眼进行同种异体角膜移植;实验4组24只,左眼进行高危(角膜碱烧伤后)同种异体角膜移植。术后第1、3、7、14天各组分别处死6只兔,进行大体观察、裂隙灯、组织学、电镜检查及免疫组化测定LYVE-1、VEGFR3及CD4的表达情况。结果(1)空白对照组在术后不同时间LYVE-1、VEGFR3和CD4均无表达。(2)实验1、2、3组:LY-VE-1、VEGFR3和CD4分别于术后第1天未出现表达,第3天出现表达,第7、14天表达增高;(3)实验4组:LYVE-1、VEGFR3和CD4分别于术后第1天开始出现表达,第3、7、14天表达逐渐增高。(4)实验1、2、3组术后第3、7、14天LYVE-1、VEGFR3和CD4表达均高于空白对照组。(5)实验4组术后第1、3、7、14天LYVE-1、VEGFR3和CD4表达均高于空白对照组。(6)实验3组较实验1、2组术后14天LYVE-1、VEGFR3和CD4表达增高。(7)实验4组较实验1、2、3组术后同时期LYVE-1、VEGFR3和CD4表达均增高。结论角膜移植术后存在CL,且在术后2周内随时间延长逐渐增多,CL可能直接参与角膜移植术后排斥反应。
Objective To study and investigate the expression and significance of LYVE-1 and CD4 in corneal lymphangiogenesis ( CL) after corneal alkali burn and corneal transplantation in New Zealand rabbit, to further demonstrate the existence of corneal lymphangiogenesis and to open out its effort and relationship with restoration ofcorneal alkali bum injury and rejection reaction after high-risk corneal transplantation. Methods 96 healthy rabbits are raised. They are di- vided into five groups randomly, taking the right eyes of the experimental group 1 as the black control group, which has no surgery; has 24 ; Models of corneal alkali bum in left eyes of 24 rabbits in experimental group 1 were established; experi- mental group 2 has 24, suture the grafts after drilling the left cornea ; the experimental group 3 has 24, all the left eyes have done allograft keratoplasty. High-risk corneal transplantation was made to left eyes of 24 rabbits in experimental group 4 after 14 days of corneal alkali burn. After the 1st day, 3rd days ,7th days and 14th days each group are sacrificed six rabbits, and take the experimental group 1 left and right corneas and experimental group 2,3 and 4 left corneas to carry on the inspection under the naked eye, the microscopy, the HE staining ,Electron microscopy and the immunity group of LYVE-1 ,VEGFR-3 and CD4 expression. Results (1 ) LYVE-1 ,VEGFR3 and CD4 have no expression after operation in black control group. (2 ) LYVE-1 ,VEGFR3 and CD4 in experimental group 1,2 and 3 have no expression during the 1 st day after operation, have expression during the 3rd day after operation and have strong expression during the 7th and 14th days after operation. (3) LYVE-1 ,VEGFR3 and CD4 in experimental group 4 have expression during the 1st day after operation and have strong expression during 3rd day ,the 7th and 14th days after operation. (4) LYVE-1 ,VEGFR3 and CD4 in experimental groupl,2 and 3 during the 3rd, 7th and 14th days after operation have higher expression than those in blank comparison group. (5 )LYVE-1 ,VEGFR3 and CD4 in experimental groulM during the 1st day ,3rd, 7th and 14th days after operation have higher expression than those in blank comparison group. (6)LYVE-1 ,VEGFR3 and CD4 in experimental group 3 during the 14th days after operation have higher expression than those in experimental group 1 and 2.7 ,LYVE-1 ,VEGFR3 and CD4 in experimental group 4 have higher expression than those in experimental group 1,2and 3 at the same time after oper- ation. Conclusion Corneal lymphangiogenesis is present after corneal transplantation, and increases gradually in the early two weeks which may participate in allograft rejection directly.
