放射损伤对小鼠骨髓间充质干细胞成骨潜能及造血干细胞龛位的影响 被引量:3

Effects of irradiation on the osteogenesis potential of bone marrow mesenchymal stem cells and the hematologic stem cells niche

摘要 目的观察放射对小鼠骨髓间充质干细胞(bone marrow mesenchymal stemcells,BMMSCs)体外成骨潜能及体内骨髓造血干细胞(hematologic stemcells,HSCs)龛位的影响。方法分离、培养正常及接受4 Gy放射后不同时间点小鼠BMMSCs,用碱性磷酸酶(alkaline phosphatase,ALP)染色法和实时定量RT-PCR检测成骨分化相关基因,鉴定BMMSCs体外成骨分化潜能的改变,并通过骨组织形态学和免疫组织化学的方法检测放射后小鼠体内成骨细胞及纺锤形的、表达神经钙黏附分子的成骨细胞(spindle-shaped N-cadherin-expressing osteoblasts,SNOs)数量的变化。结果全身照射(total body irradiation,TBI)后早期小鼠BMMSCs的成骨潜能增强,但随后迅速降低;TBI后28 d小鼠骨髓中成骨细胞和SNOs的数量均显著减少。结论4 Gy放射损伤后远期小鼠BMMSCs的成骨潜能显著降低,体内HSCs龛位也明显缩小。 Objective To investigate the effects of irradiation on the osteogenesis potential of bone marrow mesenchymal stem cells (BMMSCs) and the niche of hematopoietic stem cells (HSCs) in vivo. Methods At the different time points after 4 Gy irradiation, BMMSCs were isolated from male C57BL/6 mice and cultured. Their osteogenesis potentials in vitro were evaluated by alkaline phosphatase staining and osteogenesis-related gene expression was examined by real-time RT-PCR. Bone histomorphometric analysis and immunohistochemical staining of the spindle-shaped N-cadherin-expressing osteoblasts (SNOs) were performed to detect the changes in HSCs niche in vivo. Results Significant impairment of osteogenic differentiation of BMMSCs up to 28 days after irradiation was observed preceded by transitory enhancement at the early time after irradiation. Meanwhile, the number of osteoblasts as well as SNOs in vivo decreased greatly. Conclusion Irradiation resulted in damage in osteogenesis potential of BMMSCs accompanied by noticeable shrinking of HSCs niche in vivo.
