摘要
以芍药为研究对象,建立芍药高效的遗传转化再生体系,试验筛选最佳结果为:以茎段为外植体,用HgCl2消毒15min,基本培养基为1/2MS,抗坏血酸(100mg·L-)1,诱导愈伤阶段的植物生长调节剂(PGR)配比为ZT(2.0mg·L-1)+IAA(0.4mg·L-)1,胚状体分化阶段不加植物生长调节剂PGR;生根阶段为IAA(0.4mg·L-)1。抗性筛选时适宜的卡那霉素Km浓度为4mg·L-1,遗传转化受体为胚性愈伤组织。
The research chosed Paeonia lactiflora Pall. as object in order to build a genetic transformation regeneration system with great efficiency, The result was as following: the optimal explant was stalk, and the optimal sterilized time was 15 min. The basic medium was 1/2 MS medium with 100 mg·L^-1 vitamin C. The optimum bud induction medium was 1/2 MS and the PGR with 2.0 mg·L^-1 ZT and 0.4 mg·L^-1 IAA. The embryoid split medium was 1/2 MS medium without PGR. The rooting medium was 1/2 MS medium and PGR with 0.4 mg·L^-1 IAA. The Km density was 4 mg·L^-1 during resistance sifting. Genetic transformation receptor was germ callus.
