摘要
目的探讨自动蛋白印迹仪在抗可提取核抗原(ENA)多肽抗体谱的检测质量和性能。方法使用相同的免疫印迹法试剂盒,在同一条件下,分别用自动蛋白印迹仪和手工法检测血清抗ENA抗体。用自动蛋白印迹仪对已知阴性和阳性的血清标本各重复测定10次,以评价仪器检测的批内差异。同时用已知阴性和阳性标本各1份交替排列,各测5次,观察仪器是否存在标本间交叉污染情况。结果使用自动蛋白印迹仪进行重复检测10次的结果均一致,阴性和阳性标本之间检测结果无交叉污染,仪器与手工法的检测结果一致。结论自动蛋白印迹仪检测抗ENA多肽抗体重复性好,无交叉污染,可供临床实验室使用。
Objective To explore the quality and performance of the autoblot system on detection of antiextractable nuclear antigen (ENA) polypeplide antibody. Methods Under the identical condition, the serum levels of anti-ENA polypeplide antibody were detected by autoblot system method and manual method with the same immunoblotting kit respectively. The each positive and negative serum samples were detected 10 times repetitively by autoblot system for evaluating the intra-assay variation of the instrument. Simultaneously, the positive and negative samples arranged alternately were detected respectively 5 times with the instrument in order to observe the crosscontamination across sample. Results The results of autoblot system detection 10 times were consistent. No crosscontamination across sample was found in the positive and negative samples arranged alternately. The result of the instrument was consistent with that of manual method. Conclusions The autoblot system, with good repeatability, no cross-contamination, could be used for detection of the anti-ENA polypeplide antibody in the clinical laboratory.
