摘要
目的观察米诺环素(minocycline)对衰老痴呆大鼠学习记忆和脑组织eNOS、iNOS表达的影响,探讨米诺环素对衰老痴呆脑保护作用机制。方法Wistar大鼠随机分组:假手术组(S组)、痴呆模型组(M组)、米诺环素治疗组(MT组)。酶联免疫吸附法和免疫组织化学法检测大鼠脑组织eNOS、iNOS的含量,行为学检测大鼠记忆学习改变。结果MT、M组与S组大鼠前后两次跳台实验、水迷宫实验的结果有显著性差异(P<0.01),MT组与M组大鼠前后两次跳台实验、水迷宫实验的检测结果有显著性差异(P<0.01)。MT组iNOS表达较M组降低(P<0.01),MT组eNOS表达较M组增高(P<0.01);MT组eNOS、iNOS表达较S组增高(P<0.01);M组eNOS、iNOS表达较S组显著增高(P<0.01)。结论米诺环素能降低衰老痴呆大鼠脑组织iNOS、增强eNOS表达抑制氧化应激反应发挥脑保护作用。
Objective To investigate the effect of minocycline on the expressions of eNOS and iNOS in brain of aged dementia rat, and study its neuroprotection mechanism for aged dementia. Methods Wistar rats were randomly divided into sham, aged dementia model and minocycline treatment groups, eNOS and iNOS were measured by immunohistochemistry and ELISA. Learning memory was measured by praxiology. Results Shuttle box and morris water maze tests in minocycline treatment and aged model groups were more significantly different than those of sham group (P 〈0. 01 ). Shuttle box and morris water maze tests in minocyeline treatment group were more significantly different than those of sham groups (P 〈 0. 01 ). The expression of iNOS in minocycline treatment group was lower and the expression of eNOS was higher than that of model group ( P 〈 0. 05 ). The expressions of eNOS, iNOS in minocycline treatment and model groups were higher than those of sham group ( all P 〈 0.01 ). Conclusions Minocycline can decrease the expression of iNOS and increase the expression of eNOS in aged dementia. Minocycline can play such a neuroprotection role in aged dementia rats by restraining oxidative stress action.
