摘要
为了研究葡萄糖调节蛋白78(Grp78)对细胞迁移和粘附的影响,应用Grp78反义寡核苷酸(Grp78AS-ODN)处理人胚肾细胞HEK293,对细胞的迁移和粘附特性进行分析。结果发现应用Grp78反义核酸处理细胞可以抑制HEK293细胞的迁移,促进细胞与底物间的粘附。进一步研究发现,应用Grp78反义核酸处理细胞可以促进粘着斑的形成,并引起小GTPaseRhoA在细胞膜和细胞浆中的重新分布。上述结果表明,特异性下调Grp78的表达可以抑制细胞迁移,促进细胞与底物间的粘附。
To explore the effect of Grp78 on cell migration and adhesion, we treated HEK239 cells with Grp78 AS-ODN to knock down the expression of Grp78, and analyzed the status of cell migration and cell adhesion. We found that Grp78 knockdown inhibits the migration of HEK293, and enhances the adhesion between cells and substrate. Further study revealed Grp78 AS-ODN treatment promotes the formation of adhesion plaque, and causes the redistribution of GTPase RhoA on cellular membrane and in cytoplasm. Taken together, specific down-regulation of Grp78 could inhibit cell migration and enhance adhesion between cell and substrate in HEK293 cells.
