摘要
目的:探讨核转录因子NF-kB和基质金属蛋白酶-9(MMP-9)在甲状腺乳头状癌中的表达及意义。方法:用免疫组化SP法研究不同甲状腺组织中NF-kB、MMP-9的表达,其中甲状腺乳头状癌48例、甲状腺腺瘤18例、腺瘤旁正常组织15例。结果:甲状腺乳头状癌组织中MMP-9、NF-kB的表达明显高于甲状腺腺瘤和正常甲状腺组织(P<0.05);同时两者与甲状腺乳头状癌临床分期及颈淋巴结转移有关,而与年龄、性别无关;且甲状腺乳头状癌中NF-kB和MMP-9表达存在正相关关系(P<0.05)。结论:MMP-9、NF-kB对甲状腺乳头状癌的发生、发展有促进作用;对两者的联合检测可作为病情监控指标,有利于估计患者的预后。同时以MMP-9和NF-kB为中心的靶向治疗,可能成为抑制肿瘤转移的有潜力的途径之一。
Purpose: To investigate the expression and clinical significance of nuclear factor kappaB (NF-kB) and matrix metalloproteinase-9 (MMP-9) in thyroid papillary adenocarcinoma (TPA). Methods: We investigated the expression of MMP-9 and NF-kB in thyroid normal tissue, thyroid adenoma and TPA by SP immunohistochemical technique. TPA 48 cases; thyroid adenoma 18 cases, and 15 cases of normal thy- roid tissues taken from around the thyroid adenoma. Results: The MMP-9 and the NF-kB expression in TPA is significantly higher than that in the thyroid adenoma and the normal thyroid tissues (P〈 0.05); the MMP-9 and the NF-kB expression rate in thyroid papillary carcinoma does not correlation with age and gender, while there is significant correlation with the pathological stage and the existence of lymph node metastasis. There was significant positive correlation between the NF-kB expression and the MMP-9 expres- sion (P〈0.05). Conclusions: MMP-9 and NF-kB may play a role in accelerating proliferation in the occur- rence and development of the TPA. It is possible to use a joint test of MMP-9 and NF-kB in identifying the progress of the disease and providing a prognosis. The research also shows that a treatment plan with a focus on the level of these two items could be a promising method in inhibiting the metastasis of the tumor.
