摘要
以生长旺盛的天蓝刺头生长点为外植体,以附加不同浓度的BA、IAA、IBA、NAA、2,4-D、GA的1/2 MS或1/3 MS为培养基,进行了天蓝刺头生长点生长、不定芽分化、试管苗生根、试管苗移栽与扦插和试管苗移植的研究。结果表明:1/2MS+BA 0.2 mg/L+NAA 0.1 mg/L是芽生长的理想培养基;1/2MS+AgNO31.2 mg/L+BA 0.9 mg/L+NAA 0.1 mg/L是天蓝刺头分化培养的理想培养基;1/3MS+IAA 0.3 mg/L是天蓝刺头试管苗生根培养的理想培养基;炉灰渣和河沙是试管苗移栽和扦插的理想基质;移植于山坡上的试管苗生长旺盛,根系发达。
The growing points of Echinops dissectus were cultured on the 1/2 MS or 1/3 MS medium supplemented with different concentrations of BA, IAA, IBA, NAA, 2,4-D and GA for growth of callus induction, bud differentiation and rooting. The results showed that the optimum medium for callus induction, hud differentiation and rooting is 1/2 MS+ BA 0.2 mg/L+ NAA 0.1 mg/L, MS+ AgNO3 1.2 mg/L+ BA 0.9 mg/L+NAA 0. 1 mg/L and 1/3 MS+IAA 0.3 mg/L respectively. The optimum substrates for plantlet transplanting are slag and river sand. The transplanted plantlets in the field grow vigorously.
