摘要
目的:探讨老年高龋和无龋人群牙菌斑中变形链球菌临床分离株基因型与龋病的关系。方法:将79例老年人分为高龋组(55例)和无龋组(24例),从两组个体口腔牙菌斑中分离鉴定出263株和102株变形链球菌(血清型c),用细菌DNA提取盒提取细菌染色体DNA,经多次实验自行确定AP-PCR反应条件,对获得的变形链球菌(血清型c)临床分离株进行遗传型分析。结果:两组同一个体临床分离株共发现了101种不同基因型。高龋组同一个体口腔中定植不同基因型菌株的数量与无龋组比较差异有统计学意义(P<0.01)。结论:不同个体分离出的变形链球菌菌株间具有明显的遗传多态性;个体携带不同基因型菌株的数量与其致龋性有密切关系。
Objective:To analyze the relationship between genetic diversity of the clinical isolates of Streptococcus mutans in elderly patients with and without dental caries. Methods:Isolates of Streptococcus mutans( serotypc c)were obtained from 79 elderly patients. 263 and 102 isolates of streptococcus mutans( serotype c) were obtained from 24 caries-free and 55 caries-active patients. Chromosomal DNA was extracted from those isolates. We established the right Arbitrarily Primed Polyerase Chain Reaction(AP-PCR) reaction model for the strains. We compared the genomic differences between isolates from caries-free and those from caries-active patients in genotype analysis. Results: A total of 101 genotypes of streptococcus mutans( serotype c)strains were obtained. In caries-active patients more than one genotype isolates of streptococcus mutans ( serotype c) were colonized. The difference was statistically significant ( P 〈 0.01 ). Conclusion : Isolates of Streptococcus mutans( serotype c)obviously feature genetic diversity. The genotypes of isolates might correlate the best to differences in caries susceptibility.
