双水杨酸酯对2型糖尿病大鼠肾脏病变的作用 被引量:3

摘要 目的:2型糖尿病及其并发症的发生同炎症密切相关,水杨酸类抗炎药物可以改善胰岛素抵抗,降低血糖。双水杨酸酯结构中缺少乙酰基,没有引起消化道出血的不良反应,并且容易在小肠水解吸收。文中探讨双水杨酸酯对2型糖尿病大鼠糖脂代谢及肾脏病变的影响。方法:取72只Wistar大鼠造2型糖尿病模型后随机分为对照组、胰岛素组(INS)、双水杨酸酯组(SAL)、双水杨酸酯+胰岛素(SAL+INS)治疗组。分别在第4周和第8周时,每组各取9只大鼠,测定血糖、血脂、血肌酐、血尿素氮及24 h尿蛋白含量(TP),取肾组织作病理切片,用免疫组化方法观察肾小管上皮细胞转化生长因子-β1(TGF-β1)的表达。另外第4周时测定对照组和SAL组的三酰甘油和总胆固醇。结果:4周和8周后SAL组的血糖、肌酐、总胆固醇、TP明显降低(P<0.05)。4周和8周后INS组的血糖明显降低(P<0.05)。4周和8周后SAL+INS组的血糖明显降低(P<0.05),8周后肌酐、TP明显降低(P<0.05),8周后大鼠肾脏的病理变化明显改善。结论:双水杨酸酯能改善2型糖尿病大鼠的糖脂代谢及肾脏病变。 Objective:Type 2 diabetes and its complications are closely associated with inflammation. Salicylate, anti-inflammatory drugs, can improve insulin resistance and lower blood glucose. Salsalate, due to lack of acetyl, has no adverse reactions as gastrointestinal bleeding and is easy to hydrolysis and absorb in the small intestine. This study is to investigate the effects of salsalate on the glucose and lipid metabolism and the kidney in type 2 diabetic rats. Methods: Seventy-two diabetic Wistar rats were randomly divided into 4 groups: control, insulin (INS), salsalate (SAL) and salsalate + insulin (SAL + INS). At the end of the fourth and eighth week, blood glucose, blood lipid, serum creatinine, blood urea nitrogen and 24-hour urine protein content (TP) were measured in 9 rats of each group. Meanwhile, the kidneys were collected for pathological analysis and the transforming growth factor-β1 (TGF-β1) ex- pression in tubular epithelial cells was detected by the immunohistochemical method. At 4 weeks, blood triglyceride and total cholesterol of the control and SAL groups were measured in addition. Results: Blood glucose ( P 〈 0.05 ), creatinine ( P 〈 0.05 ), TP ( P 〈 0.05 ) and total cholesterol ( P 〈 0.01 ) of the SAL group and blood glucose (P 〈0.05 ) of the INS and SAL + INS groups were reduced significantly as compared with the controls. Creatinine ( P〈0.05 ) and TP (P 〈 0.05 ) of the SAL + INS group were decreased significantly and the pathological changes in the kidneys were improved remarkably at 8 weeks as compared with the controls. Conclusion:Salsalate can improve glucose and lipid metabolism and kidney pathological changes in rats with type 2 diabetes.
