摘要
利用花生离体茎尖作外植体,在诱导生根培养基(MSB+0.2 mg/LNAA)中分别添加20、40、60、80和100 g/L的PEG6000,统计外植体不定根的再生率,观察其生长情况,探讨培养基中添加PEG6000对花生不定根再生及生长特性的影响。结果表明:培养至第4周时,外植体在PEG浓度为60 g/L^100 g/L的培养基的生根速度明显低于对照培养基。PEG浓度为80 g/L时,外值体不定根的再生率显著降低,且品种间差异显著。渗透胁迫对不定根的生长也有明显的影响,PEG浓度为60 g/L^80 g/L时,单个外植体的主根数显著降低,且品种间差异显著;PEG浓度为40 g/L时,单个外植体长度为4~6 cm的主根数显著降低且品种间差异显著。在80 g/LPEG渗透胁迫的生根培养基上,6个供试品种的相对生根率存在显著差异,相对生根率由大到小依次为:鲁花11、丰花1号、丰花2号、鲁花12、0616和白沙1016。因此,可以根据培养4周时,80 g/LPEG渗透胁迫下外植体的相对生根率,60 g/L^80 g/L PEG渗透胁迫下外植体的主根数以及40 g/LPEG渗透胁迫下单个外植体形成长度为4~6 cm的主根数作为评价指标,利用花生茎尖离体培养鉴定花生品种间抗旱性的差异。
Adding different concentrations of PEG6000 to culture medium inducing adventitious root formation, in vitro shoot tips from peanut are used as explants to calculate rooting speed and rate of regenerated adventitious roots. Taking culture medium that doesnt contain PEG as the control, growth characteristics was investigated to compare effects of PEG on root induction, root growth , and rooting rate of different cultivated varieties of peanuts on 80 g/LPEG osmotic stress was also compared. The study shows fourth week, on medium containing 60 g/L to 100 g/L PEG, rooting that of the control. When concentration significant difference between varieties. of PEG is 80 g/L, rooting rate that, when explants are cultivated to the speed of explants decreases obviously than of explants decreases significantly and has PEG has obvious effects on growth characteristics of roots. When eoncen- tration of PEG is 60 g/L and 80 g/L, average number of main roots decreases significantly and has significant difference between varieties. When concentration of PEG is 40 g/L, average number of main roots of whieh length is between 4 to 6cm , has significant difference between varieties. On medium containing 80 g/L PEG, relative rooting rates of explants from 6 kinds of peanut materials have significant difference and the resistance to osmotic -stress from high to low is as follows: Luhua 11, Fenghua 1, Fenghua 2, Luhua 12, Baisha 1016,0616. Therefore, when explants are cultivated to the fourth week, relative rooting rate under 80 g/L PEGosmotic stress, average number of main roots under 60 g/L to 80 g/L PEG osmotic stress, average number of main roots under 40 g/L PEG osmotic stress, of which length is between 4 to 6cm, can be used as the technical indexes to identify drought resistance of peanuts with in vitro shoot tips.
