摘要 目的探讨Notch1受体和其下游信号分子Hes1、Hes5在人脑星形胶质细胞瘤中的表达及其相关性。方法应用免疫组织化学方法检测62例人脑星形细胞瘤和7例正常人脑组织中Notch1和Hes1、Hes5表达情况。结果星形细胞瘤组Notch1、Hes1和Hes5的表达均显著强于正常脑组织组(P<0.05)。在各级星形细胞瘤中,Notch1和Hes1的表达水平Ⅱ~Ⅲ级显著高于Ⅳ级(P<0.05),Hes5的表达在2组间差别无统计学意义(P>0.05)。在星形细胞瘤组中Notch1的表达与Hes1一致性较好(k=0.503),与Hes5的表达一致性差(k=0.216)。结论Notch1表达与星形细胞瘤的发生有关,与星形细胞瘤的病理分级无关。Notch1可能主要是通过下游分子Hes1而非Hes5发挥作用。 Objective To investigate the significance of the expression of Notch1,Hes1,Hes5 and their relationships in human astroeytomas. Methods Expressions of the Notch1, Hes1 and Hes5 protein in 62 human astrocytoma tissue specimens and 7 normal human brain samples were examined by immuno- histochemical method. Results The expressions of Notch1, Hes1 and Hes5 in human astrocytomas group were considerably higher than those in normal cerebral tissues group(P〈0.05). The expressions of Notchl and Hesl in astrocytomas of grade Ⅱ -Ⅲ were higher than those in astrocytomas of grade Ⅳ (P〈0.05), while there were no statistically significant differences in Hes5 expression level between two grades. There were good corelations between the expressions of Notch1 and Hes1 (k=0. 503), while the consistency between Notch1 and Hes5 was poor(k=0. 216). Conclusion Notch1 is involved in the tumorigenesis of human astrocytomas, but its expression level is not correlated with the pathological grade of astrocytoma. The role of Notch1 in astrocytoma may function through its downstream effector Hes1 but not Hes5.
