摘要
目的探讨抑癌基因IL-24对宫颈癌Hela细胞株增殖和凋亡的影响。方法将携带有目的基因IL-24的穿梭质粒pAd track CMV转染入体外培养的宫颈癌Hela细胞,用MTT法检测细胞的增殖情况,用流式细胞仪测定细胞周期及凋亡率。同时以转染空载体pAd track CMV的Hela细胞和野生型Hela细胞作为空载体对照和阴性对照。结果经MTT法测定,三组之间OD值总体上有差异(P<0.001),IL-24组OD值明显低于空质粒组和阴性对照组(P<0.001),而空质粒组和阴性对照组的OD值无明显差异(P=0.661);经流式细胞仪测定,三组之间的细胞早期凋亡率总体上有差异(P<0.05),IL-24组细胞早期凋亡率明显高于空质粒组和阴性对照组(P<0.05),而空质粒组和阴性对照组的细胞早期凋亡率无明显差异(P>0.05);与空质粒组和阴性对照组相比,IL-24组G0/G1期细胞增多(P<0.05),S期细胞减少(P<0.05),而空质粒组和阴性对照组无明显差异(P>0.05)。结论IL-24对宫颈癌Hela细胞有抑制增殖、诱导凋亡的作用。
Objective To investigate the effect of tumor suppressor gene IL - 24 on the growth and apoptosis in cervical cancer cell line Hela. Methods The shuttle vector pAd track CMV that carried the objective gene of IL- 24 was transfected into Hela cells. Effects of IL - 24 gene on growth and apoptosis of the transfected cells were evaluated by Methyl thiazolyl tetrazolium (MTT) rapid photocolorimetric assay and flow cytometry (FCM). At the same time, pad track CMV was transfected into Hela cells as empty - vector control while wild - type Hela cells as negative control. Results As showed by MTT assay, the OD values of three groups have a total difference. (P〈 0. 001). Compare to the other two groups, the OD value of IL- 24 group was significantly lower (P〈0. 001), but rio significant difference was detected between empty-vector group and negative control group (P=0. 661). As indicated by FCM, the apoptotic index of the three groups have a total difference (P〈0.05). The apoptotic index of IL-24 group was significantly higher than that in the other two groups (P〈0. 05), but no significant difference was found between empty- vector group and negative control group (P〉0. 05). In addition, compare to the other two groups, the number of cells in G0/G1 period was increased and cells in Spefiod was decreased (P〈0. 05) in IL- 24 gourp, but no significant difference was shown between empty - vector group and negative control group (P〉0. 05). Conclusion IL - 24 gene can inhibit the growth and induce apoptosis of cervical cancer cell Hela.
