摘要
目的:利用Ⅶ型胶原酶构建大鼠脑出血模型,为脑出血研究寻找理想的造模方法.方法:使用健康雄性200~250 g Wistar大鼠,采用立体定位注射的方法向大鼠脑内尾状核部位缓慢注入含0.6 U胶原酶Ⅶ的溶液1 L,术后观察大鼠行为学改变,于术后3d和7d分别取材观察血肿形成情况.结果:术后24h大鼠即有明显行为学改变,3 d可以形成直径3 mm左右的血肿,至7 d血肿区神经细胞大量坏死.结论:Ⅶ型胶原酶定位注射可以建立理想稳定的脑出血动物模型.
Objective: To establish the animal model ofintracerebral hemorrhage (ICH) with collagenase VII for the ideal model oflCH. Methods: Experimental ICH was induced by the stereotaxic injection of 0.6 U collagenase VII in 1 μL solution into caudate nucleus of rats. Then obesrved the behavior changes of model rats; 3d and 7d after the operation, take histological examinations of brain tissue respectively. Results. There were obvious hehavior changes 24h after the operation. 3mm diameter hematoma apeared 3d after the operation and a number of neuron necrosis appeared after 7d. Conclusion: ICH model established by stereotaxic injection ofcollagenase VII is stability and reliability.
