摘要
[目的]为通过紫外线诱变育种的方法来提高海产单胞藻饵料蛋白质含量提供参考。[方法]采用波长为253.7 nm的紫外线,分别辐照盐藻、三角褐指藻以及湛江等鞭藻3种海洋微藻0、2、4、6、8、10 min,测定蛋白质含量,分析紫外线辐照对3种海洋微藻蛋白质含量的影响。[结果]紫外线辐照后,盐藻的蛋白质含量与对照相比,呈下降趋势, 由对照的6.70 mg/g,最低下降到2.32 mg/g;三角褐指藻经2~10 min照射,蛋白质含量均呈上升趋势,其中辐照时间2 min的处理上升幅度最大, 蛋白质含量升高到5.28 mg/g;湛江等鞭藻经过2、4、6 min的照射,蛋白质含量分别是对照的653%、737%和153%,明显增加,而经过8~10 min的照射,湛江等鞭藻死亡。[结论]紫外线辐照对不同藻类的蛋白质含量具有不同的效应。
[ Objective ] The reference for the improvement of protein content of single cell marine algae as feed was provided through UV-induced mutation breeding method. [ Method ] The Dunaliella sp. ,Phaeoclactylum tricornutum and Isochrysis galbana Zhanjiang etc were irradiated by the ultraviolet with the wavelength of 253.7 nm for 0, 2, 4, 6, 8 and 10 minutes, respectively and the effect of the treatments on their protein content was analyzed through the deter ruination of their contents. [ Results ] Compared with the CK, the content of protein in Dunaliella sp. appeared a downward trend(6.70 mg/g to 2.32 mg/g) after UV irradiation. The content of protein in Phaeodactylum tricornutum was increased in the treat- ment from 2 to 10 minutes, among which the treatment of 2 minutes' irradiation promoted the largest content of protein( up to 5.28 mg/g) ; the content of protein in Zhanjiang and other lsochrysis galbanas that were treated for 2, 4 and 6 minutes were 653%, 737% and 153% significantly more than that of the CK and the irradiation of 8 - 10 minutes cansed their death. [ Conclusion ] The effect of UV irradiation on the protein content of different species was different.
