摘要
目的观察脑震荡(CC)大鼠囊泡乙酰胆碱转运体(VAChT)的变化规律,进一步了解CC大鼠认知行为障碍的神经生物学基础.方法自制金属单摆闭合性脑损伤复制仪复制CC大鼠模型,随机分为6个实验组:伤后ld、2d、4d、8d、16d和24d组(n=8),另设正常对照组(n=8).动物定时经左心腔灌注固定取脑,石蜡包埋连续切片,用抗VAChT抗体进行免疫组化染色,对前额叶皮质(PFC)、尾壳核脑室旁(CPu-1)、尾壳核远离脑室外侧区(CPu-2)、伏隔核(AcbC)、斜角带垂直臂核(VDB)、斜角带水平臂核(HDB)、梨状皮质(Pi)r、顶叶联合皮质(PtA)、海马(fCA1-3)、丘脑背内侧核(MDM)、丘脑背外侧核(LDVL)、下丘脑外侧区(LH)、皮质后外侧杏仁核(PLCo)等15个脑区VAChT阳性表达物进行光密度测定.结果大鼠CC后VAChT阳性表达呈下降改变,各脑区达低谷的时间各不相同,至伤后24d均恢复或接近正常,它在神经元胞体和轴突中呈颗粒状着色.结论(1)VAChT在正常大脑广泛区域的神经元中有表达;(2)CC鼠伤后VAChT表达下调可能与CC鼠的认知行为障碍有一定的关系.
Objective To observe the changes of vesicular acetylcholine transporter (VAChT) after cerebral concussion (CC) in rats in order to understand the neurobiology mechanisms of cognitive behavioral deficits after cerebral concussion (CC). Methods The cerebral concussion was induced by a metallic pendulum-striker concussive device. Then these rats were randomly divided into six injured groups: 1, 2, 4, 8, 16 days and 24 days groups (n = 8 in each group). Additionally, 8 normal rats were selected as control group. Rats were perfused through left chambers heart, the brain tissues were embedded by paraffin, then serial sections were cut. The expression of VAChT protein in the fourteen areas namely, PFC, CPu-1, CPu-2 , AcbC, VDB, HDB, Pit, PtA, CA 1-3, MDM, LDVL, LH and PLCo were studied by using anti-VAChT antibody for the immunohistochemistry, and then we detected optic density. Results The expression of VAChTin" the encephalic region above-mentioned declined compared with the control after CC. But different structure reached the lowest point and began to recovery in a quite different time. By the 24d after injury the VAChT expression has recovered to the normallevel. Conclusions (1) The positive expression matter of VAChT distribute in neurons of vast brain area. (2) The VAChT protein down-regulates after CC, this changes may be involved in cognitive behavioral deficits in CC rats.
