摘要
目的测定健康人全血中Li、Zn、Cr、Cu、Fe、Mn、Ni V、Ca、Cd和Mg共11种元素的含量。方法采用电感耦合等离子体发射光谱仪(ICP-OES),健康人血样经硝酸-高氯酸(体积比为4:1)浸泡过夜,次日置电热板上140℃消解处理后测定。结果线性范围分别为:0.50×10^-2 ~0.50mg·L^-1(Ni)、1.0×10^-1 ~4.0mg·L^-1(Li、Mn、V)、0.10×10^-2 ~0.10×100mg·L^-1(Cr、Cd)、0.10×10^-1~0.20×10^2mg·L^-1(Cu)、0.10~0.50×100mg·L^-1(Zn)、1.0~1.0×10^2mg·L^-1(Fe、Ca、Mg),相关系数均大于0.9995。回收率在83.0%~107.0%之间,精密度RSD均小于9.8%。结论本方法适合健康人全血中Li、Zn、Cr、Cu、Fe、Mn、Ni、V、Ca、Cd和Mg的含量测定,同时为进一步研究微量元素与各种疾病的关系奠定了基础。
Objective To determine eleven inorganic dements include Li, Zn, Cr, Cu, Fe, Mn,Ni, V, Ca, Cd and Mg in human blood by inductively coupled plasma optical emission spectrometry. Methods The blood was dipped in HNO3-HC1O4 (V: V = 4:1 )to stay overnight, then the samples were digested in electric heating plate set at 140 ℃. Results Assay was linear over the rang 0. 005 - 0. 500 mg · L^-1 for Ni, 0. 001 -4. 000 mg·L^-1 for Li,Mn and V,0. 001 - 10. 000 mg·L^-1 for Cr and Cd,0. 01 -20. 00 mg·L^-1 for Cu, 0. 1 -50. 0 mg·L^-1 for Zn. All of the correlation coefficients are no less than 0. 999 5. The recovery rates are between 83% and 107%. The RSD of precision are less than 9.8%. Conclusions This method can be used to determine the contents of Li, Zn, Cr, Cu, Fe, Mn, Ni, V, Ca, Cd and Mg. It provides a basis for the further study on the relationship between trace elements and health problems.
