摘要
目的探讨华蟾素对体外培养的子宫内膜癌Ishikawa细胞形态、增殖、凋亡、周期分布及侵袭能力的影响。方法体外培养Ishikawa细胞,不同浓度的华蟾素干预后,光镜观察细胞形态改变,采用MTT法检测细胞的增殖,流式细胞术分析细胞凋亡及其周期,用Transwell小室检测细胞体外侵袭能力。结果华蟾素作用后细胞由梭形、多边形变成圆形,浮起。华蟾素能有效地抑制Ishikawa细胞的增殖,降低侵袭能力,诱导细胞凋亡,并使细胞周期阻滞于G1期,减少S和G2期的细胞。华蟾素终浓度为3.0mg/ml抑制作用最明显。结论华蟾素可抑制Ishikawa细胞增殖,降低侵袭能力,诱导细胞凋亡并阻断细胞周期。
Objective To investigate the effect of cinobufacini on the biological behaviour of Ishikawa cells of endometrial cancer cultured in vitro. Methods The shape, proliferation, apoptosis, cycle and invasion of the Ishikawa ceils cultured in vitro were detected with microscope, flow cytometry and MTT colorimetry when interventioned with different concentrations of cinobufacini. Results Cells changed from polygon into round. Cinobufacini effectively inhibited the proliferation and invasion of Ishikawa cells,induced cell apoptosis, increased cell population in phase G1 and decreased in S and G2 phase, especially at the concentration of 3.0 mg/ml einobufacini. Conclusion Cinobufacini could inhibit cell proliferation and invasion, induce apoptosis and growth retardation of Ishikawa cells.
