摘要
目前对猪肉品质评价的蛋白质组学研究较少,本研究目的在于建立分辨率高、重复性好的猪骨骼肌样品双向电泳技术。应用双向电泳(2-DE)技术,优化样品裂解液组成、肌肉组织研磨方式、水化方式及等电聚焦参数等影响猪骨骼肌2-DE分离效果的因素。研究结果表明,提高猪骨骼肌总蛋白双向电泳的分辨率和重复性的方法:①液氮研磨;②8mol/L尿素、2mol/L硫脲、4%CHAPS的裂解液成分;③主动水化和被动水化相结合的水化方式;④等电聚焦采用渐进式升压方式。通过优化后的2-DE体系,建立基于双向电泳的猪骨骼肌蛋白样品制备及等电聚焦方法,为猪肉品质评价的蛋白质组学研究提供基础数据。
There is few research on the use of proteomic in the evaluation of pork quality at present, the aim of this research is to establish the pig skeletal muscle sample two-dimensional electrophoresis (2-DE) technique which has the high resolution and good repeatability. 2-DE was utilized to compare and optimize the influence factors including grinding method, hydration models and IEF parameter. So the high resolution and good repetition results are obtained,which are liquid nitrogen grinding, sample lyses solution of 8 mol/L urea,2 mol/L thiourea and 4% CHAPS, rehydration model of active hydration for 6 h and passive hydration for 6h, isoeleetric focusing method of keeping voltage increasing gradually. Thus, this article established the pig skeletal muscle proteomic method for sample preparation based on 2-DE and IEF, and provided the foundation for the future pork study through the optimized protocol of 2-DE.
