肺癌细胞系中p120-catenin亚型1A和亚型3A对E-cadherin和β-catenin的调节功能

P120-catenin Isoforms 1A and 3A Differently Affect the Expression of E-cadherin and β-catenin in Lung Cancer Cells

摘要 背景与目的p120-catenin(p120ctn)作为catenin家族的重要成员,具有多种功能各异的亚型。由于这些亚型存在着组织和细胞学特异性,因而在不同的组织和细胞当中,p120ctn各亚型可能对细胞粘附发挥不同的调节功能,但至今尚缺乏直接相关的实验证明。方法选取p120ctn低表达的肺癌细胞系A549和NCI-H460,分别向其中转染p120ctn亚型1A和p120ctn亚型3A的cDNA质粒,应用RT-PCR、Western Blot、Transwell检测p120ctn各亚型对E-cadherin/β-catenin复合体的影响,及其对肺癌细胞侵袭和转移能力的调节作用。结果p120ctn亚型1A的高表达能够通过促进E-cadherin/β-catenin的表达,进而明显抑制肺癌细胞的侵袭能力;而p120ctn亚型3A的高表达却没有这样明显的调节作用。结论P120ctn亚型1A和亚型3A可能通过对E-cadherin/β-catenin的不同调节作用,在不同程度上调控肺癌细胞的侵袭能力。 Background and objective Different isoforms ofp120-catenin (p120^ctn), the members ofcatenin family, are variably expressed in different tissues and cells. These isoforms may confer different properties with respect to cell adhesion and migration, but this concept needs experimental evidence. Methods To more precisely define their biological roles, we stably transfected isoforms 1A and 3A into the lung cancer cell lines AS49 and NCI-H460, which had low expression of p120^ctn. Using RT-PCR, Western Blot, and Transweil, we testified their impact on the expression of E-cadherin, β-catenin, and the invasiveness of lung cancer ceils. Results Expression ofisoform 1A ofp120^ctn upregulated E-cadherin and β-catenin, as well as the ability of cells to invasion. In contrast, overexpression of isoform 3A had a smaller influence on invasion. Conclusion p120^ctn isoforms 1A and 3A may differently regulate the invasive, adhesive of lung cancer cells through distinct regulation of the expression of E-cadherin and β-catenin.
