摘要
【研究目的】磷脂脂肪酸(PLFAs)是一个良好的生物标记物,可应用于土壤微生物群落多样性的定性定量分析。为提高研究分析效率,编写了生物标记辅助分析程序PLFAEco,提供基于PLFAs的微生物群落分析功能。【方法】基于MIDI公司的Sherlock脂肪酸微生物鉴定系统,采用Perl为编程语言。【结果】程序PLFAEco可计算样品的微生物群落特性,包括Shannon-Wiener(H1)、Brillouin(H2)、McIntosh多样性指数(H3)、丰富度指数(S)、Pielou均匀度(J)、Simpson优势度指数(D)计算,执行聚类分析,主成份分析;计算对象可分别为样品或PLFAs;也可依用户设定以PLFAs组合代替单一PLFA为计算单元便于比较真菌、放线菌、细菌等微生物类群之间的关系;根据样品重复、处理信息进行均值和标准差计算;计算结果以包含图表的网页形式显示,提供CSV格式的数据文件供其它软件使用。【结论】程序PLFAEco扩展了Sherlock系统在微生物群落分析方面的功能,避免了手工利用该系统带来的磷脂脂肪酸数据提取、数据矩阵构建、统计计算等复杂而繁重的工作。具有操作方便、速度快的特点,能满足大多数情况下的需求,已在烟田土壤、发酵床养猪场垫料的微生物群落分析中得到了验证。程序及手册、示例位于网站www.dagri.org。
[OBJECTIVE]Phospholipid fatty acids (PLFAs) were widely used as the biological mark to analysis the microbial community diversity qualitatively and quantitatively. Program PLFAEco was developed to analysis the PLFAs of the soil samples. [METHOD] Based on the Sherlock MIS(MIDI Inc.), the program was written with Perl script. [RESULTS] The function of program PLFAEco included: calculation of the Shannon-Wiener (H1), Brillouin (H2) and McIntosh (H3) diversity index, the abundance (S), Pielou evenness (J) and Simpson index (D); calculation of the Mean and SD; analysis of Cluster and PCA and output the statistics graphs. Output was formatted into a HTML webpage included the data table and graph, and the files (CSV format) were provided for data exchange with other software. [CONCLUSION]PLFAEco took short time to complete PLFAs data analysis and was the powerful tool for data process attaching to the MIDI system. It had been used to analysis the flue-cured tobacco soil and the stroma cushion of non-pollution pigsty. User can download the program, manual and the example data from www.dagri.org.
