牡蛎钙补肾中药血清对骨形成蛋白-4诱导成骨信号转导机制的调控作用 被引量:10

摘要 目的:用血清药理学方法研究牡蛎钙补肾中药血清对离体大鼠成骨细胞中骨形成蛋白-4诱导成骨信号转导机制的调控作用。方法:采用多次胶原酶消化法获得足够数量的成骨细胞(OB),用改良钙钴法测定碱性磷酸酶(ALP)活性以鉴定OB。用含药血清进行细胞培养48h后,用逆转录多聚酶链反应技术(RT-PCR)检测各组OB中BMP-4和Smad5、6mRNA表达。结果:与模型空白组比较,牡蛎钙补肾中药组可明显上调OB中BMP-4和Smad5 mRNA表达,下调Smad6mRNA表达。结论:牡蛎钙补肾中药血清可以提高OB中BMP-4和Smad5基因表达、降低Smad6基因表达,这可能是其防治骨质疏松症的重要机制之一。 Objective: To observe the regulating effect of serum containing oyster calcium and kidney-reinforcing Chinese herbs on signal transduetion induced by BMP-4 in rat osteoblasts (OB). Methods: OB were isolated via collagenase digestion, and identified through detecting the activity of alkaline phosphatase (ALP) with refined Gomori method. After 48h culture with the drug serum, the mRNA expression of BMP-4, Smad5 and Smad6 were detected with RT-PCR in each group. Results:Compared to the model group, BMP-4 and Smad5 mRNA were increased and Smad6 mRNA was decreased in treatment group. Conclusion:Serum containing oyster calcium and kidney-reinforcing Chinese herbs can increase the expression of BMP-4 and Smad5 mRNA and decrease the expression of Smad6 mRNA in OB, which might be one of the mechanisms for prevention and cure of osteoporosis.
