姜黄素对人卵巢癌SK细胞增殖活性及PCNA、P53 mRNA表达的影响 被引量:3

摘要 目的探讨姜黄素抗卵巢癌SK细胞增殖活性的作用及机制。方法培养好的人卵巢癌SK细胞,分别加入10、20、30、40、50μmol/L姜黄素,对照组加等量溶剂二甲基亚砜(DMSO),分别孵育24、48 h,应用MTT方法和细胞计数方法测定SK细胞生长抑制率。40μmol/L姜黄素孵育48 h后,收集细胞,RT-PCR方法测定PCNA和P53 mRNA的表达。结果姜黄素对SK细胞增殖具有明显的抑制作用。在10~50μmol/L浓度范围内,姜黄素对SK细胞增殖的抑制率随浓度增高和时间延长而增加。40μmol/L姜黄素处理SK细胞48 h后,PCNA mRNA表达显著下降(P<0.01),P53 mRNA表达显著上升(P<0.01)。结论姜黄素可通过下调PCNA表达、上调P53 mRNA表达,发挥其抑制卵巢癌SK细胞增殖的作用。 Objective To investigate the effects of curcumin on proliferation of human ovarian cancer SK cell. Methods The SK cells were cultured with curcumine (10, 20, 30, 40, 50μmol/L) or DMSO for different times (24 h,48 h), and the rates of growth inhibition were tested by cell counting and MTT assay. Cells treated with 40μmol/L curcumine for 48 h were collected and subjected to RT-PCR assays for the expression levels of PCNA and P53 mRNA. Results The results of MTT and cell counting showed that treatment with curcumine resulted in significant reduction of SK cell proliferation in a dose dependent (10,20,30,40,50 μmol/L) and time dependent (24,48 h). The RT-PCR results demonstrated that the expression of PCNA was declined regulated, and P53 mRNA expres- sion increased markedly at 40μmol/L curcumine stimulation for 48 h ( P 〈 0.01). Conclusion The curcumine could inhibits the SK ceil proliferation through down regulating the expression of PCNA and up regulating the expres- sion of P53.
