摘要
本研究以糖尿病大鼠切口伤及海绵死腔伤为难愈伤口模型,就PDWHF在改善难愈合伤口中的作用进行了探讨。致伤当日及以后连续6日,糖尿病治疗组切口伤及海绵给予PDWHF20μl(5mg/ml),伤后5、7、10、14日,测定伤口抗张力值及海绵中总DNA、羟脯氨酸含量。结果显示:伤后5~10日,PDWHF能明显改善糖尿病伤口的愈合状况,伤后14日可使愈合程度恢复至正常水平。提示无需进一步纯化。
This study was designed to inspect the accelerating effect of PDWHF(Platelet derived wound healing factors)on the impaired healing of wounds in diabetic rats.44 SD rats were divided into two groups,the normal(n=12)and the diabetic(n=32).Diabetes in rats were in duced by alloxan.Two 4cm parallel incision of full thickness rat dorsal and two pieces of polyvinyl alcohol(PVA) sponge implanted in the same rat dorsal subcutaneous pockets were selected as wound model.Injection of 20μl of PDWHF preparation(5mg/ml)into one piece of the sponge in diabetic rats was performed once a day from the injury on for 7 consecutive times,while 20μl of BSA(5mg/ml) into the other sponge in diabetic rats and into sponges in the normal rats in the same way.Meanwhile,20μl paraperasion of PDWHF/5% collagen was given to one of the incisions in diabetic rats and 20μl paraperasion of BSA/5% collagen to the other incision in diabetic rats and those in normal rats.The wound breaking strength and levels of total DNA and hydroxyproline were determined on the days5,7,10 and 14 after woundng.The results showed:PDWHF could significantly improved the impaired healing of diabetic rats on the postwounding days 5,7,l0.On the day 14 after wounding,PDWHF could completely reverse the retarded healing of wounds in diabetic subjects.
