摘要
目的:探讨人端粒酶逆转录酶(hTERT)启动子调控Bax基因在端粒酶阳性的腺样囊性癌细胞-SACC-83中表达的特异性,以及Bax对SACC-83细胞增殖的影响。方法:脂质体介导pACTERT-Bax质粒载体转染SACC-83和端粒酶阴性的人胚肺成纤维细胞(HEL),通过RT-PCR检测Bax表达,MTT法检测Bax基因表达对细胞增殖的影响,流式细胞仪检测细胞凋亡率。结果:hTERT启动子调控Bax基因在SACC-83细胞中明显表达,对该肿瘤细胞的增殖有明显的抑制作用(相对活性58.90%),且诱导细胞凋亡比例增加(11.62%);而在HEL细胞中hTERT启动子未能诱导Bax基因表达,对该细胞的增殖和凋亡无明显影响。结论:hTERT启动子可以诱导Bax基因在SACC-83中特异性表达及诱导细胞凋亡,同时还能消除Bax基因对正常细胞的潜在毒性。
OBJECTIVE: To investigate the expression specificity of gene Bax driven by human telomerase reverse transcriptase (hTERT) promoter in adenvid cystic carcinoma cells (SACC-83) with telomerase activity and the growth inhibitory capability of Bax to SACC-83. METHODS: After transfecting pACTERT-Bax plas- mid into SACC-83 cells and HEL cells through liposome, its effect on these ceils was investigated in vitro by using RT-PCR technique, MTT method and flow cytometry. RESLUTS: The expression of extrinsic gene Bax driven by hTERT promoter was detected in SACC- 83 cells with telomerase activity, and not detected in HEL cells without telomerase activity. After transfection of pACTERT-Bax, the proliferation of SACC-83 cells showed a significant inhibitory effect (the relative cell viability was 58.90 % ), and its apoptotic rate was promoted (11.62%), whereas no inhibitory effect was observed in HEL cells and its apoptotic rate showed little change. CONCLUSION: hTERT promoter can be used to induce tumor-specific expression of Bax and apoptosis in SACC-S3 cells, and avoid potential toxicity of Bax to normal somatic cells.
