液-质联用串联质谱检测干血滤纸氨基酸水平的影响因素 被引量:12

摘要 目的分析液-质串联质谱检测干血滤纸片氨基酸水平的影响因素,寻找消除这些影响因素的方法。方法设立3个影响因素组:①不同打孔位置组;②4℃条件下,不同滤纸片保存时间组;③室温条件下,不同放置时间组。比较在上述条件下检测干血滤纸片的氨基酸水平。结果不同的打孔位置对于19种氨基酸的测定没有明显的影响。不同的纸片保存时间对多数氨基酸的测定没有明显的影响,但是Glu、Gly、Pro、Ser 4种氨基酸的水平随着纸片保存时间的不同而发生了明显的变化;不同的放置时间对多数氨基酸的测定没有明显影响,但是Arg、Gln、Glu、Lys、Tyr 5种氨基酸的水平随着放置时间的不同而发生了明显的变化。结论在使用串联质谱法检测干血滤纸片上的氨基酸水平时应使用新鲜纸片,及时进行预处理,从而保证结果的稳定性和准确性。
