摘要
目的通过观察电针对排尿频率、脑桥蓝斑酪氨酸羟化酶(TH)表达的影响,探讨针刺抑制左旋多巴诱发的大鼠膀胱机能亢进的神经生物学机制。方法动物随机分成对照组、模型组、针刺组,其中模型组和针刺组动物腹腔内先后注射卡比多巴和左旋多巴引起大鼠膀胱机能亢进;在动物清醒和自由活动状态下,应用连续膀胱测压技术动态观察电针中膂俞穴对排尿频率的影响。应用免疫组织化学的方法,观察电针后脑桥排尿中枢蓝斑TH的动态变化,并用图像分析技术进行定量分析。结果模型组在腹腔注射左旋多巴后排尿频率迅速增加(P<0.05),针刺组在腹腔注射左旋多巴后15~75min时段排尿频率较处理前增加(P<0.05),75~105min时段排尿频率接近处理前水平(P>0.05)。针刺组腹腔注射左旋多巴后3h时TH在脑桥蓝斑核内的表达较对照组显著减少(P<0.05)。模型组腹腔注射左旋多巴后8h时TH在脑桥蓝斑核内的表达较对照组显著增加(P<0.05);针刺组腹腔注射左旋多巴后8h时TH在脑桥蓝斑核内的表达较对照组减少(P<0.05)。结论电针可降低左旋多巴引起的膀胱机能亢进大鼠的排尿频率;电针可抑制其脑桥排尿中枢TH的表达。
Objective To explore the neurobiological mechanism of acupuncture inhibition of L-dopa inducded rat bladder hyperactivity by investigating the effect of electroacupuncture on micturition frequency and the expression of tyrosine hydroxylase (TH) at pontine locus ceruleus. Methods Rats were randomly allocated to control,model and acupuncture groups. In the model and acupuncture groups of rats,Carbidopa and subsequent L-dopa were intraperitoneally injected to induce bladder hyperactivity. In the conscious and free-moving rats,the effect of Electroacupuncture at point Zhonglushu(BL 29) on micturition frequency was investigated by continuous cystometry and the dynamic change in pontine locus ceruleus TH was examined by immunohistochemistry and quantitative image analysis after Electroacupuncture. Results In the model group,micturition frequency increased quickly after intraperitoneal injection of L-dopa (P〈0.05). In the acupuncture group,micturition frequency increased during 15~75 min after intraperitoneal injection of L-dopa as compared with pretreatment (P〈0.05) and approached the pretreatment level during 75~105 min (P〉0.05). Pontine locus coeruleus TH expression decreased obviously in the acupuncture group at 3h after intraperitoneal injection of L-dopa as compared with the normal group (P〈0.05). Pontine locus coeruleus TH expression increased obviously in the model group at 8h after intraperitoneal injection of L-dopa as compared with the normal group (P〈0.05). Pontine locus coeruleus TH expression decreased in the acupuncture group at 8h after intraperitoneal injection of L-dopa as compared with the normal group (P〈0.05). Conclusion Electroacupuncture can reduce micturition frequency and inhibit TH expression at pontine micturition center in rats with L-dopa induced bladder hyperactivity.
