摘要
目的研究α-细辛脑的抗惊厥、抗癫痫作用机制。方法预先给予小鼠α-细辛脑腹腔注射,30min后制作癫痫模型,对小鼠脑部的ATP酶、抗氧化指标及氨基酸含量的变化进行研究,探讨其抗惊厥、抗癫痫作用机制。结果α-细辛脑能明显降低谷氨酸/γ-氨基丁酸的比值,增强抗氧化能力及ATP酶的活力,与模型组比较差异有统计学意义(P<0.01)。结论α-细辛脑抗惊厥、抗癫痫的作用机制可能与其清除自由基,维持小鼠大脑中的兴奋/抑制系统及Na+、K+、Ca2+、Mg2+的动态平衡有关。
Aim To study the anticonvulsive and antie-pileptic mechanism of α-asarone. Methods α-asarone was intraperitoneally injected (ip) in mice and acute epileptic mouse models were made after 30 min. Change of ATPase, index of antioxidation, and variation of amino acid (AA) contents in brain of epileptic mice were used to investigate α-asarone's anticonvulsive and antiepileptie mechanism. Results For α-asarone treated epileptic mice, when compared with model group, glutamate/gamma-aminobutyric acid ( Glu/GABA) was greatly decreased (P 〈 0. 01 ), capability of antioxidation and the vitality of ATPase were greatly increased ( P 〈 0. 01 ). Conclusions Anticonvulsive and antiepileptic mechanism of α-asarone may be relat- ed to its scavenging free radicals and maintaining the balance of Na^+, K^+ , Ca^2 + , Mg^2+ and excitation/inhibition systems in mice brain tissue.
