SA与高温交叉处理对葡萄叶片蛋白激酶活性的影响

Influence of Salicylic Acid and High-temperature Overlapping Processing on the Activity of Protein Kinase in Grape Leaves

摘要 以葡萄品种克瑞森无核(Vitis vinifera L.cv.Crimson Seedless)二年生扦插苗为试材,喷施水杨酸(Salicylic Acid,SA)浓度为150μmol/L后,在45℃高温下处理30 min后取样,并以喷施蒸馏水后45℃处理为对照(CK),采用体外底物磷酸化方法对叶片中蛋白激酶的活性进行了测定。结果表明,交叉处理和对照的叶片中蛋白激酶在底物髓鞘碱性蛋白(myelin basic protein,MBP)浓度为0.5 mg/mL时,其活性分别达到最大值10028.19和7 843.26 cpm,但是交叉处理比对照高2 184.94cpm。而后随底物浓度的进一步增加,其活性下降。Mg^2+浓度为5 mmol/L时,交叉处理和对照的叶片中蛋白激酶活性也分别达到最大值10 285.15和7425.22 cpm,但交叉处理明显高于对照,然后,随着Mg^2+浓度的升高两者的激酶活性而明显地降低。Ca^2+对蛋白激酶活性的影响很弱,Mn^2+对蛋白激酶活性的激活没有影响。ATP浓度为50μmol/L时,交叉处理和对照的叶片中蛋白激酶的活性分别达到了最大值10 108.43和7 921.48 cpm,然后随着ATP浓度的增加蛋白激酶的活性不断下降。当反应时间达到10 min时,交叉处理和对照的叶片中蛋白激酶的活性分别达到了最大值9 979.41和7 248.63cpm,随着时间的增加,蛋白激酶的活性开始迅速下降。 The experiment was conducted to examine the effect of salicylic acid and high - temperature overlapping treatment on the activity of protein kinase in grape leves. Two - year - old grape plants Crimson Seedless( Vitis vinifera L. )cuttings were used as experimental materials. The salicylic acid with the concentration of 150 μmol/L was sprayed and the treatment at 45℃ for 3 min was carried out after spraying distilled water at 45℃ as CK. and then , the activity of protein kinase in grape leaves after treatment was measured through substrate phosphorylation in vitro. The results showed that the activity of protein kinase in leaves of the overlapping processing and CK reached a peak value 10 028.19 cpm and 7 843.26 cpm as MBP concentration was 0.5 mg/mL, which is 2 184.94 cpm high than CK, and then the activity of protein kinase decreased slightly as MBP concentration increased continuously. The activity of protein kinase in leaves of the overlapping processing and CK exhibited a maximum value10 285.15 cpm and 7 425.22 cpm as Mg^2 + concentration was 5 mmol/L in reaction system with MBP substrate, but the overlapping processing is continuously higher than CK, and then the activity of protein kinase decreased obviously with the increase of Mg^2+ concentration. Effect of by Ca^2 + on the activity of protein kinase was extremely weak. Mn^2 + had no stimulative effect on the activity of protein kinase. The activity of protein kinase in leaves of the overlapping processing and CK reached a peak value 10 108. 43 cpm and 7 921.48 cpm, as ATP concentration was 50 μmoL/L, then, the activity of protein kinase decreased gradually as ATP concentration increased continuously . When the reaction time was 10 min, the protein kinase activity in leaves of the overlapping processing and CK reached the maximum 9 979.41 cpm and 7248.63 cpm , as time increased. protein kinase activity began to decline rapidly.
