摘要 [目的]建立烈香杜鹃叶总黄酮含量测定的RP-HPLC方法,并比较它们的含量。[方法]烈香杜鹃叶用甲醇提取,反相高效液相色谱法分析药材中槲皮素、山奈酚、异鼠李素3种黄酮甙元,并计算出总黄酮含量。[结果]采用Kromasil C18柱(4.6 mm×250 mm),以甲醇-0.4%磷酸(60∶40,V/V)为流动相,可使3种黄酮成甙元达到基线分离,平均加样回收率98.74%和98.98%,相对标准偏差1.11%和1.00%。[结论]RP-HPLC法可作为总黄酮的定量分析方法。 [ Objective ] The research aimed to establish RP-HPLC method for the determination of flavonoids in leaves of Rhododendron athopogonoide Maxim and compare their contents. [ Method ] Quercetin, kaempferol and isorhamntin in leaves of Rhododendron athopogonoide Maxim were extracted by methanol and analyzed by RP-HPLC, and then the content of fiavonoids were calculated. [ Result] When the analysis was performed on a C18 column(4.6 mm × 250 mm)with methanol-0.4% phosphoric acid(60:40, V/V) as mobile phase, the quercetin, kaempferol and isorhamntin were separated well, and the average recoveroes were 98.74% and 98.98% , RSD were 1.11% and 1.00%. [ Conclusion] RP-HPLC could be used for quantitation determination of flavonoids.
