摘要 用蛋白激酶C(PKC)刺激剂佛波醇酯(PMA)和PKC抑制剂Rottlerin处理培养神经元,RT-PCR法检测神经元NF-кBp65mRNA的表达;用PMA和NF-кB抑制剂TPCK处理培养神经元,流式细胞术AnnexinV/PI法检测细胞早期凋亡率。观察PKC对原代培养神经元核转录因子kappaB(NF-кB)表达的影响,探讨PKC参与缺血性神经元损伤的可能机制。结果显示PKC可促进NF-кB的表达;NF-кB可诱导培养神经元的早期凋亡。由此可以得出PKC激活参与神经元的损伤可能是通过引起神经元内NF-кB的表达而实现的。 To investigate the effects of protein kinase C (PKC) on the expressions of nuclear factor kappa B (NF-B) in primary cultured neuron, and to explore the mechanism of PKC participated the ischemic injure of neuron, methods like cultured neurons treated with PMA (the PKC stimulator) and Rottlerin (a selective PKC inhibitor), the expression of NF-κBp65mRNA detected by RT- PCR and the cultured neurons treated with PMA and TPCK (a selective NF-κB inhibitor) were used. The early cell apoptosis was detected by flow cytometry stained with AnnexinV/PI. Result showed that PKC could promote the expressions of NF-κB. NF-B could induce the early apoptosis of the primary cultured neuron. It indicated that PKC could participated the injure roles of neuron through the expression of NF-κB.
