在血液与生物材料的相互作用中,血浆蛋白特别是纤维蛋白原(Fibrinogen,FG)的吸附起着重要的作用。本文利用酶联免疫吸附检测手段(ELISA),测定了两种鼠抗人FG的单克隆抗体M1和R1同吸附在材料表面的FG结合的亲合力(Ka)。由于两种抗体与FG结合的位点不同,其Ka值随温度和放置时间的变化呈现不同的变化规律,从而推测材料表面吸附的FG的构象变化。
Protein adsorption, especially, fibrinogen (FG) adsorption played a very important part in the action of blood with biomaterials. In this paper, the affinity Ka of the two mouse moloclonal anti-human FG antibodies M1 and R1 and adsorbed FG to biomaterials was determined by Enzyme Linked Immunosorbent Assay (ELISA). Because the two antibodies were baund with FG in different sites, based on the difference of the effect of temperature and residence time on Ka, the conformation changes of adsorbed to FG in the surface of biomaterials could be deduced.
