摘要
以水稻液泡膜H+-ATPase为研究对象,探讨了造成诱导性冷失活现象的机制.运用MC540荧光对诱导性冷失活过程中的膜脂状态研究表明,冷失活处理后膜脂的有序程度大幅度下降(MC540荧光发射增强56%),并且通过与常温下诱导物(Mg2+、ATP)扰动膜脂的效应比较,证明诱导物在冷失活条件下能诱导膜脂有序性迅速丧失,其后才出现酶活丧失.根据实验结果和膜脂-膜蛋白相互作用关系。
the mechanism of cold inactivation on rice tonoplast H + ATPase is studied,paying special attention on the influence of tonoplast lipid.MC540 is used to detect the packing order of tonoplast lipid,the result showed this order is destroyed in a great level(fluorescence emission increased by 56%).The same influence of the inducers(Mg 2+ ,ATP)is relatively weaker.This result demonstrated the inducers can induce the destruction of the order of tonoplast lipid,then,the enzyme activity declined.According to the above results and the lipid protein interaction,we consider that the tonoplast lipid plays an improtant role in cold inactivation of H + ATPase.
