动物性食品中磺胺二甲嘧啶残留酶联免疫吸附测定法复核实验研究

摘要 为确保公司出栏前鸡肉中磺胺二甲嘧啶残留不超标,同时减少实验室的劳动强度,提高工作效率,节约检验费用,特进行动物性食品中磺胺残二甲嘧啶残留酶联免疫吸附测定法复核试验,以确认公司是否可以用北京望尔生物技术有限公司研制的磺胺二甲嘧啶试剂盒做为出栏前的筛选试验。试验分别用酶联免疫吸附试验及液相色谱法对试剂盒的灵敏度、准确度和精密度以及与仪器方法的对比实验进行了测试。结果表明:试剂盒Icso平均值为2.78/~g/I。,波动范围在2.09~3.45μg/L之间.鸡肉最低检测限为1.07μg/kg.经测定磺胺二甲基嘧啶添加的回收率范围为73.2%~112.8%,批内变异系数在5.2%~13.4%之间.与仪器方法相比较,望尔试剂盒的准确度与仪器检测结果相同,阴阳性符合率为100%,且前处理方法简单.经验证,该方法可用于磺胺二甲嘧啶残留的筛选检测. To ensure that sulfadimidine sodium residues in company's chicken before yarding do not exceed the standards, decrease the amount of human labor needed in the laboratory, increase work efficiency and save the cost of test, inspection experiment of ELISA Method for analysis of sulfadimidine sodium residues in animal-derived food was conducted, so as to confirm that ELISA kit from Beijing Wanger Bio- logical Technology Co. Ltd can be used in the screening experiment for chicken before yarding. ELISA test and liquid chromatography were respectively used to study the sensitivity, accuracy and precision of Wanger ELISA kit. The comparison experiment between it and HPLC test was also carried out to check the consistency of the two methods. The result of the experiment showed that the average of IC50 of Wanger test detection limit in blank chicken sample was 1.07 μg/kg. The average recovery between 73.2%~112.8%, inter-lot coefficient was between 5.2%~13.4% The ELISA kit test results were close to HPLC test, and the consistency of samples were 100%. Considering the simple sample preparation, the ELISA kit screening of sulfadimidine residues. kits was 2.78μg/kg. The of all spiked samples was both negative and positive is applicable for the rapid screening of sulfadimidine residues.
