摘要
以珠帘藤带芽节段和叶片为外植体进行组织培养.结果表明,以节段外植体诱导丛芽的适宜培养基为MS+2mg/LBA,以叶片外植体诱导不定芽的适宜培养基为MS+1mg/LBA+0.1mg/LNAA;茎段外植体成芽时间较短、数量较多;最佳不定芽增殖培养基为MS+1.5mg/LKT,繁殖系数达5.5,苗壮、长势好;最佳不定芽生根培养基为1/2MS+0.1mg/LIBA+0.6g/LAC,生根率达96%,平均生根7.5条,根粗壮;最适试管苗移栽基质为腐质土+珍珠岩+椰糠(1∶1∶1),移栽成活率达92%.
Stem segments with lateral buds of Cissus sicyoides 'Ovata' or its leaves were used as explants in tissue culture. The results showed that the optimal media for inducing adventitious buds were MS medi um supplemented with 2.0 mg/L BA for stem explants and with 1.0 mg/L BA and 0.1 mg/L NAA for leaf explants. Stem explants formed more adventitious buds in a shorter time than leaf explants. The optimum multiplication medium for adventitious buds was MS medium supplemented with 1. 5 mg/L KT, which gave a proliferation rate of 5.5 and in which the shoots were vigorous. The medium 1/2 MS supplemented with 0.1 mg/L IBA and 0.6 g/L AC gave a rooting rate of over 96%, with an average root number per plantlet of 7.5. The regenerated plantlets were transplanted successfully in a mixture soil with a mud..per lite:coir dust ratio of 1: 1 : 1 with a survival rate of 92%.
