摘要
目的:探讨给孕鼠输注活化Th1细胞建立孕鼠子痫前期模型的可行性及其机制。方法:孕鼠组及未孕鼠组共接受2次Th1细胞输注。孕期检测血压以及尿蛋白,观察肾脏改变,流式细胞术分析CFSE标记Th1细胞的分布情况,并检测子宫细胞Th1/Th2细胞因子表达变化。结果:①在输注Th1细胞后孕鼠组与对照组相比其收缩压显著增高,差异有统计学意义(P<0.01);②妊娠第14天检测孕鼠24 h尿蛋白定量为(1.54±0.56)g/L,与对照组相比差异有统计学意义(P<0.01);③HE和PAS染色显示,输入Th1细胞孕鼠组出现肾脏病变;④CFSE标记Th1细胞在血液、蜕膜及肾脏中均有分布,其荧光强度值以蜕膜中最高;⑤输注Th1细胞的孕鼠子宫淋巴细胞TNF-α以及IL-12的表达率较输注PBS的孕鼠显著提高,差异有统计学意义(P<0.05)。此外子痫前期小鼠子宫淋巴细胞IL-4和IL-10的表达率显著提高。结论:输注活化的Th1类细胞可以诱导孕鼠特异性的产生典型的子痫前期样症状,通过这一方法建立的小鼠模型是合适的子痫前期模型。其发生机制与母胎界面Th1/Th2细胞因子平衡紊乱有关。
Objective: To explore the feasibility and mechanism of animal model of preeclampsia established by the infusion of Thl ceils. Methods: The rats in pregnant group and non - pregnant group both received infusion of activated Th1 cells twice. Blood pressure and urine protein were detected in pregnancy and renal changes were observed. CFSE labeled Thl cells were analyzed by flow cytometry to describe the distribution of Thl cells, and the alteration of Th1/Th2 cytokines expression in uterus was inspected. Results: (1)After the infusion of activated Thl cells, the systolic pressure of pregnant group was significantly higher than that of non - pregnant group (P 〈 0. 01 ) . (2)On the fourteenth day in pregnancy, there was significant difference in urine protein level between pregnant group and non - pregnant group ( P 〈 0. 01 ) . (3)Obvious histological changes in renal were found in Th1 infused pregnant rats. (4)Th1 cells were distributed in blood, decidua and renal. The mean fluorescence intensity in decidua was higher than those in blood and renal. (5)NF - α and IL - 12 levels of Thl cells infused pregnant rats were higher than those of PBS infused pregnant rats (P 〈0. 05) . Furthermore, IL -4 and IL - 10 levels of preeclampsia rats were higher than that of control group (P 〈 0. 05 ) . Conclusion: Infusion of activated Thl ceils can induce typical symptoms of preeclampsia in pregnant rats, which is an appropriate model. The mechanism is related to unbalance of Th1/Th2 on maternal-fetal interface.
