黄连素预处理对大鼠心肌细胞凋亡蛋白表达的影响 被引量:6

摘要 目的探予黄连素预处理对大鼠心肌缺血再灌注时心肌细胞凋亡蛋白表达的影响。方法60只SD大鼠随机分为假手术组、缺血再灌注组及黄连素预处理高、中、低剂量组。黄连素预处理组分别给予高、中、低剂量(0.1、0.2、0.4mg/kg)黄连素腹腔注射,连续7d,末次于冠脉结扎前2h腹腔注射给药。假手术组和缺血再灌注组腹腔注射等量生理盐水。结扎部位位于左冠状动脉前降支中上1/3处,结扎1h后剪开结扎线,用止血钳夹闭胸腔,再灌注2h,采用免疫荧光法检测凋亡蛋白Bcl-2、Bax的表达。结果与假手术组相比,缺血再灌注组Bax和Bcl-2阳性表达增强;黄连素各组与缺血再灌注组相比,Bax表达量降低,Bcl-2表达量上升,Bax/Bcl-2比值下降,且各剂量组间指标值有显著差异。结论黄连素预处理对心肌缺血再灌注损伤大鼠具有抑制心肌细胞凋亡的作用,且该作用具有量-效关系。 Objective: To investigate the effects of berberine preconditioning on expression of the apoptotic protein of cardiac muscle in rats. Methods: 60 SD rats were divided into 5 groups randomly: sham operation group, ischemia reperfusion (I/R) group, and high, medium and low dose berberine pretreatment group(0, 1mg/kg; 0. 2mg/kg; 0.4mg/kg). The rats of each berberine pretreatment groups had been giving different dose berberine via intra peritoneal injection one time every day for 7d, while the rats of others groups had been giving equivalent normal saline. All the rats had been given peritoneal injection of those medicine separately at the last time 2h before coronary artery ligated (Location: at the middle and superior 1/3 part of anterior descending branch of rats left coronary artery) . The ligatures were opened lh later and closed rats thorax using hemostat immediately, then took reperfusion for 2h. After that, the expression of the apoptotic protein of rats cardiac muscles were detected by immunofluorescence. Results: Compared with sham operation group, the positive expression of Bax and Bcl-2 of ischemia reperfusion group was reinforced significantly. Compared with ischemia reperfusion group, the positive expressions of Bax and the Bax/Bel-2 of different dose of berberine pretreatment groups were decreased, the positive expressions of Bcl-2 increased, and there was a significant difference among different dose of berberine pretreatment groups. Conclusion: Berberine preconditioning on myocardial ischemia reperfusion injury in rats can restrain cardiac muscle apoptosis. It shows a dose-dependent relationship.
