猪圆环病毒Ⅱ型rep蛋白的亚细胞定位生物信息学析 被引量:1

摘要 从GenBank发表的PCV-2全基因组序列,随机选择其中的32个全基因序列,做序列比对和同源性比较,绘制系统进化树,在注释中找到ORF1编码的rep蛋白氨基酸序列,用在线生物信息学工具对其进行亚细胞定位分析。结果基本符合"来自同一地区的基因序列同源性较高"的规律,表明了PCV的高度保守性,亚细胞定位分析发现其rep蛋白定位于核区的指数为0.76,这与病毒在宿主细胞核内复制分不开。结果同时表明其定位于过氧化物酶体的指数显著不同于其他(过氧化物酶体平均0.36,可变,溶酶体、线粒体均为0.1不变)这可能与pcv的组织嗜性以及复制靶细胞(多为单核/巨噬细胞)的原因相关。 32 sequences of complete genome of PCV-2 from different locations which code the protein rep were randomly selected from the genbank, then sequence alignment was done to get the origine relationship of them, and the phylogenetic tree was drew,the sequence of the protein rep was selected from the note of the gene sequence, then the sequence of the protein rep was done with protein subcellular localization prediction. The result of the prediction approximately reached the rule that gene sequences from a same location are relatively more similar to each other, and it also showed that the gene of the PCV was hard to change. The result of the protein subcellular localization prediction showed that the certainty of the protein rep of the PCV-2 locating in the nucleus was 0.76, which is relative to the truth that the virus reproduce in the nucleus, but the result also showed that the index of the protein rep of the PCV-2 located in perexisome was far different from the other cell crgans( the averagy of the mlcrobody was 0.36 and variable, but the index of the mitochondrial matrix space and lyso- some was 0. 1 and stable ), this may have relationship with the truth that the PCV-2 prefers to a certain tissue and the truth that the target cell of the reproducing of the PCV-2 are more likely the monocyte and the maerophage.
