摘要
目的研究葡萄糖对大鼠血管平滑肌细胞增殖的作用与Ca^2+水平的关系。方法取SD大鼠胸主动脉,用组织贴壁法原代培养血管平滑肌细胞,实验使用第4-8代细胞,荧光酶标仪的方法检测细胞内游离Ca^2+水平。结果葡萄糖对大鼠VSMCs的增殖具有一定的浓度效应;随着葡萄糖浓度的增加,在一定范围内胞内Ca^2+浓度也相应增加;随着时间的延长,细胞内的Ca^2+浓度也逐渐升高。结论葡萄糖引起大鼠血管平滑肌细胞的增殖可能与其引起细胞内的Ca^2+浓度的升高有关。
Objective To study the effect of glucose on vascular smooth muscle cells (VSMCs) and its relationship with intracellular calcium concentration. Methods Diabetic model was induced in rats, VSMCs were separated from rat aorta and cultured with explant culture method. Cells were identified by method of immunohistochemistry. The intracellular calcium concentration of VSMCs was assayed with the method fluorescent enzyme immunoassay. Results The proliferation of VSMCs was induced by glucose in a dose-dependent manner. And the intracellular calcium concentration increased by the stimulation of glucose in dose and time-dependent manner. Conclusion The effect of glucose on VSMC proliferation might be associated with the induction of intracellular calcium concentration.
