摘要
HARDY(HRD)是AP2/ERF-like家族转录因子的一员,已有的研究表明它的过量表达可以增强水稻的抗旱性、提高水稻的水分利用效率。为了研究它是否在其他的农作物中具有普遍的适用性,本试验从拟南芥中克隆了HRD基因,成功构建了HRD基因的高效植物表达载体pBIN-HRD。采用农杆菌介导法转化番茄(亚心87-5),经PCR、RT-PCR分析表明,阳性的转HRD基因植株已获得表达。
HARDY(HRD) is a member of the AP2/ERF-like transcription factor family. Overexpression of HRD in rice improved its drought resistance and water use efficiency (WUE). In order to study whether it has a universal applicability in other crops, we cloned the HRD gene from ArabidoDsis thaliana (Columbia) genome by PCR method and successfully constructed the plant expression vector pBIN-HRD in this study. Tomato plants were transformed via Agrobacterium tumefaciens. PCR and RT-PCR analysis indicated that the foreign gene HRD had been integrated into the genome of tomato and had acquired expression. This work lays a foundation for further study on the function of drought resistance and water conservation of HRD transcription factor.
