摘要
目的研究急性缺氧对血管内皮细胞活力和凋亡的影响。方法人脐静脉血管内皮细胞,缺氧(5%O2)条件下培养0、1、3、12h,四甲基偶氮唑盐比色法(MTT法)检测细胞活力,膜联蛋白荧光素法(Annexin V-Fluos)检测相应时间细胞凋亡率。结果急性缺氧早期,血管内皮细胞的活力增加,缺氧3h达顶峰,之后随缺氧时间延长有所降低,至缺氧12h低于正常。凋亡检测结果显示,缺氧3h即能引起内皮细胞凋亡,12h凋亡进一步加重。结论急性缺氧早期细胞活力及凋亡均增强,提示细胞适应性反应与损伤性反应并存,这种适应性反应将有助于细胞对抗缺氧造成的损伤。
Objective To explore effects of acute hypoxia on vascular endothelial ceil activity and apoptosis. Methods Human vascular endothelial cell line (ECV304) was cultured in 5% CO2 incubator for 0 h,1 h,3 h,12 h. MTT method was used to detect the activity of the cultured cells, and Annexin V-Fluos method was applied to detect the apoptotic rate of the cells in restricted time. Results At the early stage of hypoxia, endothelial activity enhanced, reached the peaked level at 3 h after hypoxia, then decreased thereafter with the duration of hypoxia, but was still lower than the normal level. Apoptotic analysis revealed that 3 h hypoxia could induce endothelial apoptosis, with the apoptotic rate increasing to ( 22.64 ± 0.63 ) % , then apoptosis decreased at 12 h following hypoxia. Conclusions At the early stage of hypoxia, both cell activity and apoptosis of endothelial cells enhanced following acute hypoxia, indicating that adaptive and traumatic responses co-existed in the endothelial cells. This adaptive response would help cells to resist injuries induced by hypoxia.
