人β淀粉样蛋白基因植物表达载体的构建及转化

摘要 人β淀粉样蛋白(Amyloidβ peptide,Aβ)是治疗阿尔茨海默氏病(Alzheimer′s disease,AD)的关键靶标之一。本试验利用重叠PCR技术获得2Aβ串联基因片段,并将其克隆到植物表达载体pBIDST(含双35S启动子和TEV增强子)中。用根癌农杆菌EHA105介导转化烟草,筛选获得了卡那霉素抗性植株。PCR和RT-PCR检测结果证实2Aβ基因已经整合进烟草基因组中并进行了转录。 Human amyloid β peptide (Aβ) is one of the key targets in treatment of Alzheimer's disease. The two tan- dem fragment of Aβ obtaining by overlap PCR technique was cloned into plant expression vector pBIDST (containing dou- ble 35S promoter and TEV enhancer). The 2Aβ gene was introduced into tobacco by Agrobacterium turnefaciens EHA105. Then the kanamycin resistant tobaccos were obtained.The results of PCR and RT-PCR indicated that the 2Aβ gene was integrated into the genome of tobacco and transcribed.
